DESCRIPTION:The  International BIFI 2022 Conference will take place online from February 7th to 9th\, 2022. On this occasion the Conference will be focused on the Science of Covid-19: from molecular drug design to data-driven epidemiological models.  \nIn order to ease attendance\, this year’s conference will be entirely hosted online. \nEvery two years the Institute organizes an international conference around its research areas. This is the case of 2022 when we will be holding the BIFI 2022 International Conference which will focus around what we could call the Science of Covid-19. As we well know at this time\, the subject is highly topical and possibly expected\, but given the massive impact to our societies and its relation to the core activities of our institute\, it is also natural choice for 2022. We hope this occasion will serve to consolidate knowledge on the topic\, bring new insights and tools to it\, and also to fill some gaps and open new research paths. The Conference aims at providing a forum to discuss the latest advances and insights in the study of this pandemic. All topics that fit within the science of COVID-19 from a complexity\, biological or interdisciplinary point of view are welcome. \nOn the website of the event (http://bifi22.bifi.es/) to be announced all the necessary information including the access link in due time.

DESCRIPTION:Title: Glycochemistry and Molecular Recognition: Rational and Combinatorial Approaches \nAbstract: \nCarbohydrates participate in a large variety of molecular recognition processes of biological relevance\, involving both proteins receptors\, enzymes and nucleic acid fragments. Understanding how these later bio-molecules interact with glycosides represents a fundamental issue in chemical biology with far reaching implications in fundamental biology\, biotechnology or drug design. On the other hand\, the acknowledgment of the relevance of glycoconjugates in biology has run parallel to the development of new chemical methods for glycosidic bond formation. Thus\, the last few decades have witnessed a burgeoning progress in the area of glycosyl donor engineering\, with a more recent focus on the understanding of glycosylation mechanisms. As a part of an ongoing project focused on glycochemisty we\nhave analyzed different aspects of the association of glycosides to both nulcleic acids and proteins. In addition\, key mechanistic features of glycosylation reactions\, both in enzymatic nd chemical contexts\, have been dissected. From a methodological persepective our approach is pluridisciplinar and includes NMR\, molecular modeling\, organic synthesis\, and a variety of chemical strategies. DESCRIPTION:Title: Neurodegenerative proteinopathies: implications and molecular mechanisms of MOK signaling kinase in neuroinflammatory responses. \nAbstract: \nNeurodegenerative diseases\, including Alzheimer’s\, Parkinson’s and Amyotrophic lateral sclerosis (ALS) are incurable proteinopathies characterized by progressive neuronal loss\, aggregation and deposition of specific proteins in the central nervous system (CNS) and sustained neuroinflammation. In the last few years\, it has become increasingly accepted that such protein aggregates have a principal role in triggering neurotoxic immune responses. Still\, and particularly for ALS\, the molecular mechanisms and proteins participating in these pathogenic processes remain largely unknown. Recently\, we have identified MOK Ser/Thr kinase as a TDP-43-interacting protein in an ALS microglial cell model (Leal-Lasarte et al.\, FASEB J. 2017). DESCRIPTION:Title: Ecology of Online Social Systems \nAbstract \nOnline social networks have significantly changed global communication dynamics and\, in general\, the way we interact. Every day our brain is flooded by information coming from a plethora of sources and this cognitive bottleneck translates into hyper-competition for a scarce resource: our attention. This pressure\, in turn\, pushes information producers to mutualistically interact with specific memes and actors\, seeking the virality of their messages. Finally\, memes’ chances to persist and spread are subject to changes in the communication environment. The fact that all these terms — competition for resources\, mutualism and environmental changes– are\nreminiscent of another research field –ecology–\, suggests that\, at least at a coarse-grained level\, “information” ecosystems and natural ecosystems present strong similarities that can be exploited to understand their behavior.\nIn this talk\, building on these analogies\, I will show how theories and tools developed for the study of ecological communities help to gain insights on human behavior in online social interactions. In the first part\, I will introduce an analytical formalism –based on the neutral theory of ecology–\, capable of capturing the competition for attention and explaining several emergent patterns observed in information ecosystems. DESCRIPTION:Title: From structure to mechanisms of Zika virus induced neurodevelopmental disease \nAbstract:\nZika virus (ZIKV) infection was declared a global threat for public health due to its association with congenital neurodevelopmental birth defects\, such as microcephaly\, a rare medical condition characterized by a significant decrease in brain size. ZIKV infection delays cell cycle progression and induces cell death of neural progenitor cells (NPCs)\, leading to reduced production of neurons. Despite the enormous basic and clinical research efforts\, no specific therapy has yet been approved for the treatment or prevention of ZIKV infection\, thus a better understanding of its pathogenesis is an urgent need.\nOur recent studies of the effect of ZIKV proteins on NPC proliferation show that the protein NS5 that is essential for viral genome replication\, is also sufficient to alter the mode of division of NPCs. NS5 interacts and depletes primary cilia and centrosome components\, causing an atypical non-genetic ciliopathy and premature neuron delamination. While the Methyltransferase (MTase) and RNAdependent RNA polymerase (RdRP) activities of ZIKV-NS5\nappear to be dispensable\, the Y25\, K28 and K29 residues that are involved in NS5 oligomerization\, are essential for the localization and interactions with proteins of the cilia base\, promoting ciliopathy and premature neurogenesis. In\nthis talk I will summarize our recent findings\, exploring these unexpected interactions between ZIKV-NS5 and different cilia/centrosome proteins. DESCRIPTION:Tittle: Networks Virology. Learning from SARS-CoV-2 \nAbstract: \nViruses are reproductive machineries formed mainly by genetic material (RNA or DNA) and proteins. In general\, these proteins interact with specific proteins of the host forming vast networks of virus-host protein-protein interactions (PPI). Although a virus may infect mainly an organ or system of the host\, the damages can propagate beyond it\, transforming the affection into a multiorganic/multifunctional one. This is the case of SARS-CoV-2\, which makes COVID-19 a multiorganic disease affecting a dozen of organs/systems in humans. Here\, I develop the hypothesis based on facts that this propagation of extrapulmonary damages takes place via the PPI network of virus-host interactions. It considers that some viral proteins interact with specific human proteins highly expressed in the lungs. Then\, these perturbed human proteins navigate outside the lungs via an exosome-mediated transport network\, which allows the inter-organ cross-talk. Due to the fact that these “perturbated” proteins interact with their partners in other organs they are capable to transmit such perturbations beyond the lungs. Therefore\, there is a network of perturbators from the lungs interacting with a network of vulnerable proteins in other organs\, which trigger a range of damages in them. We identify here the perturbators and vulnerable proteins in COVID-19\, identifying 13 organs/systems that may be affected by the infection of SARS-CoV-2 and explain some of the extrapulmonary damages observed in clinics. We propose a series of drugs that can be repurposed to treat combinations of these damages in COVID-19 patients. Finally\, at a lower size-scale we zoom on these SARS-CoV-2 proteins to investigate their topological structure\, which form other networks of amino-acids interacting noncovalently among them. Instead of searching and modifying natural proteins\, de novo protein design crafts new proteins from physical principles with sequences and geometries not restricted to those seen in nature. From scratch\, protein backbones are computationally generated according to geometrical\nrestrictions defined by the designer\, followed by the calculation of low-energy amino acid sequences stabilizing these backbones. In this way\, protein\nstructures are tailored for stability and function; potentially overcoming frequent limitations of repurposed natural proteins in terms of stability\, geometry\, size\, recombinant expression or protein interactions\, among others. We have identified key principles for the de novo design of a variety of protein folds containing beta-sheets\, which are attractive structural motifs for building ligand-binding sites\, and used these principles for computationally designing proteins in excellent agreement with their experimental structures. This has opened the door to custom-design proteins for binding small-molecules and protein targets of therapeutic relevance. DESCRIPTION:Title: Direct reciprocity and model-predictive strategy update explain the network reciprocity observed in socio-economic networks \nSpeaker: Fabio Dercole \nAbstract:\nNetwork reciprocity has been successfully put forward (since M.A. Nowak & R. May’s\, 1992\, influential paper) as the simplest mechanism — requiring no strategical complexity — supporting the evolution of cooperation in biological and socio-economic systems. The mechanism is actually the network\, that makes agents’ interactions localized\, while network reciprocity is the property of the underlying evolutionary process of favoring cooperation in sparse rather than dense networks. In theoretical models\, the property holds under imitative evolutionary processes\, that however lack rationality in the socio-economic context\, whereas cooperation disappears in any network if imitation is replaced by the more rational best-response rule of strategy update. In social experiments\, network reciprocity has been observed\, though the imitative behavior did not emerge. What did emerge is a form of conditional cooperation based on direct reciprocity—the propensity to cooperate with neighbors who previously cooperated. To resolve this inconsistency\, we show network reciprocity in a model that\nrationally confronts the two main behaviors emerging in experiments — reciprocal cooperation and unconditional defection — with rationality introduced by means of a multi-step model-predictive rule for strategy update. \nSala de Conferencias\, Edif. DESCRIPTION:The IX International BIFI 2020 Conference will take place in Zaragoza\, in the auditorium of the R+D Building\, Río Ebro Campus (Actur)\, from February 3rd to 5th\, 2020. \nOn this occasion the Conference will be focused on “New Challenges in Molecular Biotechnology“. Thus\, we have prepared a scientific program with national and international speakers that we believe is very attractive. The event format includes plenary sessions with 18 talks by invited speakers\, 5 of which will be given by prominent researchers who conduct their research in renowned research centers such as the Pasteur Institute in Paris\, the Mitochondrial Biology Unit in Cambridge\, the Estación Experimental de Aula Dei (CSIC ) in Zaragoza\, the Rocasolano Institute in Madrid\, and the Laboratory of Systems Biochemistry (CIB-CSIC) also in Madrid. In addition\, nine of the works submitted will be chosen to give short oral talks of 12 min (+3 question-minutes). Two Poster sessions have been scheduled from which two works presented in this modality will be awarded with the amounts of €300 (First prize) and €200 (Second prize). \nWe warmly invite young and not so young researchers\, and also students\, to attend this new edition of our biennial conference and to send scientific communications related to the main tracks of the event. I will review the advances in cryo-EM technology that made the ¨resolution revolution¨ possible\, and then show its application to understand G-protein coupled receptors (GPCRs). GPCRs are the largest family of membrane proteins in the human body and are a target for ~35% of the currently approved FDA drugs. GPCRs sense a variety of molecules and transduce the signal to the intracellular milieu by coupling to heterotrimeric G-proteins. We used cryo-EM to obtain high-resolution structural information on GPCR-G protein complexes\, starting to tackle one of the major questions in the field – the GPCR-G protein selectivity code.
